1.研究发现，在脑胶质瘤、结直肠癌、肺癌、胃癌等肿瘤中均存在MGMT基因启动子甲基化[1-5]。其中，以在脑胶质瘤中的研究最为广泛和深入。脑胶质瘤中MGMT启动子甲基化的频率在半数以上，研究多围绕甲基化与替莫唑胺等药物治疗疗效的相关性展开[1-2, 6]。

2.药物应答方面：研究发现，MGMT启动子甲基化的脑胶质瘤患者经替莫唑胺治疗的生存期更长[1, 6]，但MGMT甲基化与PCV化疗（甲基苄肼+洛莫司汀+长春新碱）的疗效无关[7]。还有研究显示，MGMT甲基化能够降低脑胶质瘤患者经替莫唑胺后的复发率，且能延长复发时间[8]。

3.生存方面：MGMT甲基化与脑胶质瘤患者无进展生存期和总生存期均呈正相关[7]。

4.2013版美国国家癌症综合网络（NCCN）临床治疗指南建议胶质母细胞瘤患者应行MGMT甲基化检测，对于甲基化阳性患者推荐使用替莫唑胺治疗。
